Titanium alloy surface modification with polyphenolic layer and zinc ions for sustained trimethoprim delivery: a coordination-mediated antibiotic immobilization approach.

Titanium-based implants are commonly applied in orthopedic and dental treatments, yet their durability is limited by issues such as bacterial adhesion and the mechanical mismatch between the implant and natural bone. In this work the surface modification of titanium implants was investigated by developing a polyphenolic layer containing zinc ions for the sustained release of the antibiotic trimethoprim (TMP). The modification process involved the self-oxidizing polymerization of epigallocatechin gallate (EGCG) molecules on the titanium surface, with Zn2+ facilitating the formation of a coordination bond with the EGCG –OH groups. The TMP was then attached to the layer through interactions with Zn2+ ions. The modified surfaces were characterized using Scanning Electron Microscopy, X-Ray Energy Dispersive Spectroscopy, and FT-IR Microscopy. All applied techniques confirmed the effective formation of the coating and the sorption of trimethoprim. Antioxidant properties were assessed via the DPPH assay, and antibacterial activity was tested against E. coli and S. aureus strains. Additionally, biocompatibility was evaluated through cytotoxicity, cell proliferation, and osteogenic differentiation assays using hFOB 1.19 cells. The results confirmed effective modification of the titanium surface, with the material exhibiting significant antioxidant activity, sustained TMP release, antibacterial properties, and biocompatibility.
